Myelogenous Leukemia M2b

Abstract Objective: To investigate the cytogenetic features and molecular biological features of acute myelogenous leukemia M2b (AML-M2b) and the relationship between AML-M2b and t (8;21) leukemia.Methods: In patients with AML -M2b, karyotype was studied by using G-banding technique, AML1/MTG8 fusion gene transcript by using RT-PCR and the rearrangement of AML1 gene and MTG8 gene by using Southern Blot technique.Results: The frequencies of AML-M2b with or without t (8;21) were 86.1%(31/36) and 13.9%(5/36) respectively. AML1/MTG8 fusion gene transcript occured not only in all 44 AML-M2b(44/44) including 5 cases without t (8;21), but also in 12 of 13 cases during complete remission. The rearrangement of AML1 gene and of MTG8 gene was found in 81.8% and 71.0% of AML-M2b respectively.Conclusion: AML-M2b and t (8;21) leukemia is one and the same disease entity in which AML1/MTG8 fusion gene is its molecular marker.AML-M2b could be subclassified into t (8;21)(+)AML1/ETO(+)AML-M2b and t(8;21)(-) AML1/ETO(+) AML-M2b. AML1/MTG8 fusion gene transcript still may occur in AML-M2b during complete remission.
